I'm not really into "picking up" girls. Usually, all relationships start out as friendships for me, and I usually meet girls through other people and get closer to them or I'll know them from somewhere else (often school). If I had to pick up a girl though, it'd be with definte body language. If I know a girl is eyeing me, I'll eye her back obviously and although I'm not the type to really approach random girls, if she's by herself I'd most probably make a move (provided that she's eyeing me back; if she's not then screw her).

I guess I use my sense of humor, and when you talk to a girl it helps if you really listen and keep your eyes on their face. If you catch a girls eyes and shes been looking at you then its normally an invite for you to walk up to her. haha
most of the girls i meet r at house parties so i always make sure i bring some kinda of girly drink or liquor and i will pull one out so they see it and flock to me for one..it works all the time..its basically buying a drink for a girl at a bar/club.
